Administration

Thanks to its complex properties, acriflavine is the foundation of a highly versatile product – YELLOW DRINK. First synthesized in 1912, acriflavine was one of the earliest antibacterial agents used to treat infectious diseases, even before the discovery of penicillin.

It was widely used during World War I as an antiseptic and for the treatment of coma. The U.S. Food and Drug Administration (FDA) has approved acriflavine as a safe drug for topical treatment of skin lesions.

Today, acriflavine is recognized for its antiviral (including HIV), antitubercular, fungicidal, and anticancer properties. More recently, it has been proposed as a potential treatment for SARS-CoV-2 due to its activity against PLpro enzymes involved in coronavirus replication.

Its biological activity comes from its ability to efficiently bind to DNA, which allows it to disrupt various cellular functions – making it a multidirectional agent. Acriflavine inhibits HIF-1 dimerization, tumor growth, and vascular development.

HIF-1 is a key factor in cellular adaptation to hypoxia and plays a critical role in cancer progression. Cell-based screening has shown that acriflavine directly binds to HIF-1α and HIF-2α, inhibiting HIF-1 dimerization and transcriptional activity. Pretreatment of prostate cancer xenograft-bearing mice with acriflavine prevented tumor growth, while treatment of mice with existing tumors led to growth arrest. These results highlight acriflavine’s strong inhibitory effect on tumor growth and vascularization.
